The 300 square foot unit will typically be in a narrow building. Something in the 40'-wide range will work perfectly. Generally that means a former bungalow side with setbacks, not a zero-lot-line site.

In many cases it's more profitable to build 600 square foot units. The spreadsheets are extremely complex for this stuff. Much of it depends on the specific property and land use code for that spot. If you can build a bigger building with wide floorplates and one-bedrooms instead of studios, you probably will.
